Different types of bladdercancer cells share common genetic ancestry penn_state NatureComms
For single-cell RNA sequencing, tumor tissue was processed immediately after removal from the bladder. Briefly, tumor tissue was cut into small pieces were used for further analysis . Peak locations were determined using the MACS2 algorithm with a cutoff of-value = 1e−7. Peaks that were on the ENCODE blacklist of known false ChIP-Seq peaks were removed. Differentially modified H3K27ac loci were identified by the DiffBind package . Peak annotation was performed using the Homer program .
Slides were incubated with secondary antibody for 30 min at RT, washed with TBST , incubated with the Opal 520 fluorophore working solution for 10 min at RT, and washed with TBST . Slides then underwent heat-mediated antigen retrieval and antibody removal in citrate buffer. After cooling, the process was repeated for two additional rounds for labeling with anti-CD8, CD56, CYP27A1, PD-L1, and p63, followed by secondary labeling with the respective fluorophore working solution [i.

Conserved transcription factors promote cell fate stability and restrict reprogramming potential in differentiated cells - Nature CommunicationsTransdifferentiation has been proposed as an approach for regenerative medicine, but the mechanisms that safeguard cell identity are not well established. Here they identify transcription factors that oppose transdifferentiation and show that knockdown of these genes improves recovery after myocardial infarction.

P2X3 receptor antagonism attenuates the progression of heart failure - Nature CommunicationsDespite medications, heart failure worsens with time with many patients dying within five years of diagnosis. Here the authors show that blocking purinergic receptors in the carotid body stops heart failure progression, improves its function, reduces sleep apneas and systemic inflammation in male rats.

Sleep cycle-dependent vascular dynamics in male mice and the predicted effects on perivascular cerebrospinal fluid flow and solute transport - Nature CommunicationsWhy fluid and solute transport in perivascular spaces is enhanced upon sleep remains elusive. Here, the authors show that each sleep cycle state displays unique perivascular dynamics, which enhances predicted fluid movement and solute transport.
