By Neha MathurJul 12 2023Reviewed by Benedette Cuffari, M.Sc. In a recent study published in Med, researchers examine host responses to the severe acute respiratory syndrome coronavirus 2 within the placenta using transcriptomics.
There also remains a lack of research on the disproportionate COVID-19 risk burden during pregnancy including maternal illness, placental immunity, SARS-CoV-2 intraamniotic infection, and fetal disease. All maternal SARS-CoV-2-positive patients tested positive by clinical nasopharyngeal swab within 72 hours of delivery; however, not all mSARS-CoV-2+ patients had detectable viral transcripts in the placenta. All samples were collected during the SARS-CoV-2 Delta-dominant period between August 2021 and October 2021.
RNAscopy or in situ RNA hybridization was used to localize SARS-CoV-2 spike transcripts. Likewise, a one-step quantitative reverse transcriptase-polymerase chain reaction assay was used to determine the limit of detection for SARS-CoV-2 spikse, nucleocapsid protein, and open reading frame 1ab , whereas immunohistochemistry was used to stain for these viral proteins.
Visium was subsequently used to determine whether any placental spatial transcriptomic niches could be used to detect SARS-CoV-2. To this end, none of the control placentas exhibited any SARS-CoV-2 transcripts, as well as one and two placentae from asymptomatic and symptomatic mSARS-CoV-2+ cases, respectively.
