A novel tagmentation-based tool specifically predicts genome-wide off-targets of prime editors. NBThighlight

HeLa cells were seeded in a 24-well plate in preparation for transfection. One day after seeding, cells were transfected with an adequate amount of plasmid and 2 μl Lipofectamine 2000 .

1 × 10K562 cells were electroporated with the above-mentioned quantities of plasmid via a Neon transfection system . One day after transfection , antibiotic selection was conducted using puromycin at a concentration of 2 mg/ml. Puromycin selection was continued for 2 weeks [for TAPE-seq and fluorescence-activated cell sorting ], 4 weeks , or 2 days . Genomic DNA was purified with a Blood Genomic DNA Extraction Mini Kit following the manufacturer’s instructions..
